Estimation of Haemoglobin with the help of Sahli's Haemoglobinometer


Estimation of Haemoglobin with the help of Sahli's Haemoglobinometer or Acid Haematic or Visual method :


Principle: Haemoglobin is converted to acid haematin by N/10 HCL.

Materials:

  1. Sahli Haemoglobinometer: It is a Visual method. It consists of:-
                                                                     a. Matching Box
                                                                     b. Haemoglobinometer tube.
                                                                     c. Haemoglobinometer pipette
  2. N/10 HCL. 
  3. Distilled water.
  4. Stirrer.
  5. Blood collection set

Specimen: Capillary blood or EDTA anticoagulated venous blood can be used.

Procedure:

  1. Clean the haemoglobinometer tube and pipette and ensure that they are dry.
  2. Fill the haemoglobinometer tube to the 20mark with N/10 HCL
  3. Suck the capillary or venous blood in the haemoglobinometer pipette exactly up to the mark 20 µl. Wipe blood off outside of pipette.
  4. Transfer the blood from the pipette into the acid of the tube by gently blow.
  5. Rinse the pipette by sucking up acid and mix solution in tube by stirrer. 
  6. Exactly after 5 minutes, then add drop by drop Distilled water with dropper & mix well by Stirrer until the blood –acid solution matches standard. 
  7. The reading is taken and recorded in percentage and/or g/dl.
